FOR 948 P3: Degradation of chloroplast proteins- involvement of senescence associated vacuoles and cysteine proteases
Beschreibung:
The major fraction of proteins to be degraded during senescence resides in chloroplasts. So far, the proteases responsible for mass protein degradation during senescence have not been identified. It is hypothesized that cysteine proteases outside of chloroplasts are involved in degradation of chloroplast proteins and that these cysteine proteases are located in small vacuoles appearing in the cytosol during senescence (SAV=senescence associated vacuoles). In this project the mechanism of mass degradation of chloroplast proteins will be addressed by studying the formation of SAVs in senscing leaves and by characterization of protein release from the chloroplasts. To investigate the mechanism of chloroplast dis-mantling and to identify the key proteases involved in chloroplast protein degradation, the proteome of SAVs and vesicles derived from chloroplasts will be analyzed. The regulation of chloroplast protein degradation will be studied in transgenic barley plants with modified concentrations of key cysteine proteases.
