Description:
Adaptation of forage grasses to biotic and abiotic stress factors being favoured by anthropogenic climate change is a major challenge for plant breeders. The project aims at improving varieties of ryegrass (Lolium spec.) by (i) accelerated resistance gene pyramiding in the model species Lolium temulentum; (ii) marker-assisted estimation of genome proportions in backcrosses with L. perenne and L. multiflorum; (iii) metabolomic and genomic bulked-segregant methods in order to develop markers for resistance and complex traits applicable in breeding ;(iv) elucidation of resistance mechanisms against stem rust on a cellular level; (v) generation of expression and metabolic profiles and their interpretation. Setup and phenotyping of segregating populations and breeding lines will be realized by the breeding company. Characterized plant materials are then distributed to the business partners for genomic and metabolomic analyses, and in addition, fungal isolates will be delivered to the university partner for characterization of stem rust resistance. The latter task will involve laser dissection microscopy. Establishment and/or modification of molecular genetic tools (BE-STDGE, qRT-PCR) and metabolomic methods (NMR) shall entail marker development in order to support gene pyramiding and thereby accelerating gene introgression during the breeding process.
