Purpose of research: Experimental development

The detection and specific identification of plant pathogens is mainly realized by lab-based methods. Consequently, in case of a reasonable suspicion for an infestation of a plant with a quarantine organism, samples have to be sent to a specially equipped laboratory. Until the investigations are completed the trade with affected plants is stopped. Besides the time required for the analysis, high economic losses occur. Hence, the aim of the project was the development and testing of a qualified lab-on-a-chip system for the specific detection of nine important, in parts on European level regulated Phytophthora species. Therefor the reprocessing of various plant materials and the DNA extraction as well as the subsequent PCR for the amplification und labeling of single-stranded pathogen DNA and the hybridization on a biochip were optimized. In extensive studies the specificity and sensitivity of the system for nine species-specific capture probes encoding the ras related protein (Ypt1) and one genus-specific probe were demonstrated. Due to the conjunction of different process steps (PCR, hybridization, detection), a suitable laboratory prototype could be implemented.
