Purpose of research: Basic research

The evaluation of the potential for teratogenic/developmental toxicity remains a major chal¬lenge in chemical risk assessment. Established OECD guidelines are based on animal stud-ies that are preferentially performed with rats or rabbits (OECD 414 and 416). However, interspecies extrapolation of animal data to humans is difficult, since profound species dif¬ferences in respect to sensitivity towards birth defect-inducing chemicals exist (Newman et al., 1993). In addition, interindividual variability in susceptibility is to be expected within the human population. Further information from molecular mechanistic and toxi¬cokinetic studies are required to define reasons for variability between and within species and thus to improve and refine risk assessment.
