Purpose of research: Applied research

Chemical and physical parameters of soil samples and forest characteristics were assessed in an area-wide grid in the 2nd Soil Survey in Germany. The biodiversity of microorganisms in forest soils will be characterised as an accompanying investigation. The cultivation-independent methods based on DNA analyses are favourable because of the characterisation of microbial communities without the modificatory selection pressure of artificial agars. In these techniques, the 16S-rDNA of Bacteria and Archaea and the ITS1-5.8S-Its2 region of fungi are especially suitable for a taxonomic evaluation. In a first pilot study the reproducible extraction of DNA from forest soil samples with a high content of humic acids will be optimised and the T-RFLP technique (Terminal Restriction Fragment Length Polymorphism) will be established.
