Collaborative project: Development of a biological control system for the regulation of the pathogen of ash drive dying Hymenoscyphus fraxineus; Subproject 1: Screening and characterization of antagonistic microorganisms and microbial consortia to control of the pathogen of ash drive dying (Frax-ProMic)
Project
Project code: 22006116
Contract period: 01.06.2017
- 31.05.2020
Budget: 378,491 Euro
Purpose of research: Applied research
Ash drive dying fraxineus caused by Hymenoscyphus has spread since the mid-1990s across Europe and provides stocks by the death all the forest future of the ash in question. The biological control of the disease through microbial antagonists, which directly combat the harmful organism or suppress competition, is a promising option to the containment of ash drive dying and can a synergistic supplement to the Breeding of resistant genotypes represent. In the project, a biological control system will be developed to fight of the pathogen on the basis of epi - and endophytischer isolates that stable can establish itself in the ash. Through a multi-stage process, promising bacteria and fungi are genetically characterized and evaluated with regard to their antagonistic effect. Providing effective antagonistic microorganisms is to contribute significantly to the containment of ash drive dying. Perspective microbial drugs should be used in seed orchards to the development of resistant plants. In the first the micro biomes of resistant and susceptible to ash three complementary Sequenzierungsansätze (NGS of marker genes) analyzed and identified taxa or strains with antagonistischem potential. Parallel to this, a variety of bacterial and fungal strains is grown and classified (MALDI-TOF-MS). Based on the comparative analysis of resistant and susceptible to ash micro biomes (culture-sensitive and culture-insensitive) potential antagonists (isolates and microbial consortia) be selected. After the screening of the antagonistic potential in vitro (via Ko cultivation) and the examination of relevant metabolic benefits the greenhouse test of best isolates ash seedlings and the final characterisation is done their antagonistic properties.
