This project investigates molecular mechanisms underlying grapevine resistance loci against Plasmopara viticola (downy mildew). Resistance loci and candidate genes elaborated in our previous work are characterized in regard to gene expression and gene diversity. Gene-specific molecular markers will be developed for best possible application in marker-assisted breeding of new grapevine cultivars. The generation of novel grapevine cultivars with improved sustainable resistance and elite quality is a task of the research supported by the Federal Government.
Bioinformatic analyses identified candidate genes for resistance in the sequenced region of Rpv10. Sequence differences between the resistant and susceptible haplotypes of this locus could be demontrated in the genes and the functional domains of encoded proteins. This finding enables to apply molecular methods to isolate the genes and characterize their function. This work provides important basic knowledge to investigate the mechanism of resistance.
