Spraing in potatoes, caused by the Tobacco Rattle Virus (TRV), is increasingly becoming a problem in potato cultivation. The tuber deficiencies caused by the virus, such as plugging, tuber malformation and necrosis, lead to refusal of whole lots. The aim of the present project was to develop a simple, stable and sensitive diagnostic method for Tobacco Rattle Virus (TRV) in potato. The work focused on the establishment of an optimal sampling procedure, RNA isolation and the development of an isothermal detection system. The influence of TRV infection on tuber yield was determined in extensive field trials. In "TRV2GO", a sensitive rapid test for the detection of TRV infections in potato tubers was successfully established. The comparison with the previously used nested RT-PCR shows a higher sensitivity as well as a higher specificity due to the use of dipsticks.
