The spread of invasive alien species, caused by anthropogenic activities is one of the most harmful threats to biodiversity (Convention on Biological Diversity, CBD). The Asian longhorn beetle Anoplophora glabripennis (Cerambicidae) is one of these species and its early detection in infested trees is of great importance to fastly eradicate the beetle and be able to identify appropriate quarantine zones. Unfortunately the presence of exit holes does not secure the presence of this beetle, due to similarities with native cerambicid species. Other phytosanitary measures than monitoring are in most cases only justified if the adult beetle was trapped and identified using morphological and molecular techniques. In addition, the presence of exit holes does not necessarily offer the opportunity to trap Anoplophora glabripennis in one of its development stages (larvae, adult beetles). The diagnostic procedure, which ought to be developed by analyzing beetle frass and borings, intends to provide a secure tool of detection and identification of cerambicid species for the use of phytosanitary inspections, even if the organisms itself are not further present. One working package of this research proposal is to develop (in cooperation with a commercial life science supplier) a DNA-extraction Kit that uses physico-chemical extraction procedures which allow for segregation of beetle/larvae feces and skinning from wooden material. The second working package aims at developing a specific molecular diagnostic measure to finally assess and validate the previously compiled procedures.
