Purpose of research: Experimental development

The aim of this project is the evaluation of differences in the content of methyl salicylate between and within different birch species depending on the developmental stage and season regarded. Therefore, established birch cultures in different botanical gardens as well as newly cultivated plants should be investigated. Via HPLC, the content of the most valuable secondary metabolites, especially methyl salicylate and pentacyclic triterpenes like betulin and lupeol will be determined. Those compounds are described to have high antiinflammatory, antitumor, antiviral and vulnerary activity. Furthermore, non-targeted metabolic screening by UHPLC-MS7MS will investigate new and not yet described compounds. Additionally, vibrational spectroscopy methods like near infrared (NIRS), mid infrared (MIRS) and Raman spectroscopy (RS) in combination with different chemometric methods will be applied to develop qualitative and quantitative models for taxonomic discrimination and screening of the secondary metabolite profile. Hence, the project will finally offer new insight into inter- and intraspecific variation of methyl salicylate content and other valuable plant secondary metabolites of different birch species. The spectroscopy part would allow fast taxonomic classification of unknown species with special regard to optimized breeding programs.
