Purpose of research: Experimental development

The Wheat dwarfing virus (WDV) is transmitted by the leafhopper Psammotettix alienus. WDV-infection causes significant losses in yields of barley and common wheat. Because of the ban on neonicotinoids in 2018, symptoms of WDV are becoming more frequent in France, particularly in the central and north-eastern regions. The only available control method is the use of insecticides. However, their effectiveness is reduced by the high mobility of the leafoppers. Currently, no wheat or barley varieties are available for cultivation that are resistant or tolerant to this virus. For wheat and barley, some sources of partial resistance have been identified, and recently some sources of total resistance have been identified (unpublished) for barley. Within the project both cereals will be processed. For wheat, 300 accessions from Southeast Europe (a region where some tolerant accessions have already been identified), synthetic wheat (durum wheat x Aegilops tauschii) and introgression lines from several wheat-related wild species will be studied. In a broad screening approach, studies in the field, in the gauze house and in the greenhouse with partly artificial infection are carried out. The panel to be tested is genotyped in whole or in part with a high-density microarray with the aim of identifying resistance QTLs (Quantitative Trait Loci) by association genetics. Starting from accessions with pronounced resistance, bi-parental populations will be established, which could be used for mapping of resistance factors. For winter barley, a fine mapping of the already known WDV tolerance QTLs will be performed with the aim to develop molecular markers that are very close to the incriminated genes and can be used directly by breeders. As with wheat, phenotyping is carried out under controlled conditions in the greenhouse and in the field. After the third year, it is planned to study a mapping population, which will be created from recently identified sources with pronounced WDV resistance.
