Table grape cultivars and breeding lines with varying degrees of resistance to powdery and downy mildew were characterized phenotypically and genetically. Genetic finger print analysis revealed that many genotypes can be traced back to a single mildew resistant donor. About 11,000 ESTs (sequenced cDNAs) were generated from a subtractive cDNA library, consisting of cDNAs from leaves and berry skins of nine mildew resistant genotypes by subtracting V. vinifera transcripts of a susceptible cultivar. Bioinformatic examinations identified 49 ESTs with sequence similarities to resistance genes which were mapped to the grapevine reference genome. In real-time PCR analyses the origin of these ESTs could be assigned to specific Vitis wild species. Finally, two ESTs were identified which both co-localize with a resistance locus for downy mildew, encode a NBS-LRR-like resistance protein, and are originated from a wild Vitis species.
