Purpose of research: Experimental development

Xanthomonas hortorum and Ralstonia solanacearum are two bacteriosis which cannot be treated by chemicals. They are able to kill Pelargonium plants within a short time. Methods for testing resistance of both pathogens were developed at the JKI and a wide assortment of cultivated and wild Pelargoniums of the company Elsner pac® Jungpflanzen was tested. Additionally virulent strains were selected for the testing. Some resistant or tolerant genotypes were found and three of them were resistant against both pathogens. However, they cannot be crossed with cultivated varieties. Therefore they were crossed with genotypes which are crossable both with these wild types as well as with cultivated varieties. The crossing work is still in process. In order to combine these incompatible genotypes, a protocol for generation and regeneration of protoplasts was developed for different tissues and genotypes. First protoplast fusion experiments resulted in new plants which were characterized phenotypically and with various cytologic and moleculargenetic methods.
