Type IV secretion in Yersinia: Occurrence, molecular characterization and genotyping of Yersinia isolates harbouring a conjugative type IV system

The horizontal gene transfer (HGT) by conjugative plasmids is a main pathway for the dissimination of antimicrobial resistance and virulence genes in bacteria. We use the genus Yersinia as a model to study a new conjugative system discovered in Yersinia enterocolitica with close relationship to type IV secretion systems of other pathogenic gramnegative bacteria. In order to investigate if antimicrobial resistance or virulence genes are associated and transmitted by the transfer system a screening of a Yersinia strain collection was performed. The aim of this project is to elucidate resistance profiles of the Yersinia isolates and to find out if a correlation with the occurrence of the type IV system exists. Relevant resistance genes will be analyzed by molecular methods. Furthermore, fingerprint typing methods such as PFGE and AFLP are applied and improved to characterize the relationship of strains carrying the transfer system.
