Purpose of research: Basic research

Susceptibility towards xenobiotics (e.g. chemicals, additives, biozides) can be either genetically determined ore influenced by environmental factors and genetic variability may not necessarily be reflected on the phenotype level. The principle of metabolic activation represents the basis for the project: in most cases, metabolic activiation by xenobiotic metabolizing enzymes is the prerequisite for the adverse effects of xenobiotics. Xenobiotic-metabolizing enzymes vary within the human population and also susceptibility towards the adverse effects of xenobiotics varies. Therefore, the project is aimed at the investigation of whether and to which extent genetic variabilities of xenobiotic-metabolizing enzymes is also reflected at the phenotype level (e.g. protein expression, protein activity) for a better understanding of gene-environmental interactions. Modern methodologies (TaqMan technology) are applied to identify genetic variations. The results of the project will help to improve the identification of susceptible subpopulations which have to be taken into consideration in the risk assessment process. The results can be directly used in the risk assessments currently underway and also in the MOS (margin of safety)- concept. In addition, the results of the project can be included in WHO/IPCS (International Programme on Chemical Safety) activities which favour the replacement of conventionally used assessment factors (default-values) by data-derived, more realistic factors.
