Collaborative project: Development of efficient high throughput (HT) techniques for selection of grapevine cultivars with high acid stabilty, 1: Development of markers for acid content for MAS in crossbreeding of Vitis vinifera - subproject 2
Project
Project code: 2814507310
Contract period: 01.02.2011
- 31.01.2014
Budget: 222,233 Euro
Purpose of research: Experimental development
Climate change will cause major changes to viticulture in future decades, as many traditional varieties show best performance in the cool German climate. The correspondingly high organic acid production is an important factor for quality in wine production. Molecular markers linked with quality parameters would enable early selection in breeding, largely independent of environmental factors. Marker-assisted selection (MAS) could therefore result in a reduction of the selection process by up to 10 years. The project included, apart from growers, nurseries and breeders also the JKI Institute for Grapevine Breeding Geilweilerhof and the Hochschule Geisenheim University. Genomic regions important for acidity were identified in a cross population using QTL analysis at JKI. Over four years, parameters for acidity were analyzed weekly through the whole ripening period in each individual. This large phenotypic dataset enabled, together with a high-resolution genetic map, the implementation of informative QTL analysis. At Geisenheim three putative genes were identified in biologic data basis of Vitis, which largely resembled the already known gene of L-Idonatdehydrogenase (L-IdnDH), but apparently have differential functions. In nutrient solution acid production could be induced by a modified potassium supply. The knowledge generated during the project may be used in future studies to develop functional molecular markers for MAS to be applied in grapevine breeding and clonal selection.
