Purpose of research: Basic research

Polycyclic aromatic hydrocarbons (PAH) are genotoxic carcinogens which are detectable in numerous consumer products, as for example hammer grips or cable sheets. Sources of PAHs are softener oils, carbon blacks and so forth. Although some of these chemicals had long been recognised as potent carcinogens in skin carcinogenesis assays in mice, very little is known about organ-specific toxicity or penetration in human skin. However, such data are urgently required for exposure and risk assessment of these compounds, especially for PAHs that show strong genotoxic effects and/or are frequently detectable in consumer products (e.g. Benzo[a]pyren). Further, it is not yet clear whether these compounds are sig-nificantly metabolised in human skin, putative metabolites remain to be identified and char-acterised in respect to individual skin penetration and internal exposition. It is of special in-terest to analyse whether an effective barrier function of human skin is compromised by compound-mediated induction of apoptosis or inflammation.
