Purpose of research: Applied research

For genotoxicity testing of chemical ingredients of cosmetics or commodities such as apparel and others, a variety of toxicological assays have been developed. In recent years the Comet assay was propagated between laboratories. In principle, this assay is suited to investigate different organ tissues or different cell types in a sensitive and reproducible manner.In 2007, we established test protocols to reproducibly conduct the Comet assay with different keratinocyte cell lines and commercially available 3D full thickness skin models (see report 358, 2007). These protocols did not work well for the corresponding epidermis model (high background), and thus need to be adapted and further optimized in 2009.In addition, we will establish a protocol for performing the Comet assay in a highly differentiated model for bronchial epithelia (EpiAirwaysTM). Exposure of consumers to compounds present in commodities usually occurs via both dermal penetration and inhalation. In terms of genotoxicity endpoints suited to be measured in the respiratory tract, however, there is actually only little data available and the development of sensitive and reliable protocols is still on the list of things that need to be urgently addressed. The department of consumer safety at BfR is going to fill this gap in the years to come. A first step will be the development and optimization of the Comet protocol applied to human lung carcinoma epithelial cells (e.g. A549) and a human bronchial epithelia model (EpiAirwaysTM). This protocol will be established based on a few model compounds that have been successfully applied in corresponding skin models.
