Purpose of research: Applied research

Fast detection and quantification are crucial in the development of fire-blight prognosis and control strategies. Aside species-specific detection protocols, monitoring of environmental factors with impact on fire blight development is of relevance. In EUPHRESCO PHYTFIRE the partners will imply and evaluate new techniques for monitoring of Erwinia amylovora and its accompanying microflora. A new method for high-throughput microbial identification originally developed for clinical diagnosis will be applied for analysis of environmental samples. At JKI Dossenheim we will test this MALDO-TOF MS based method for population analysis of fire blight samples. We will contribute to evaluation of specific detection and on-site diagnostic techniques. Population monitoring in relation to different fire blight control strategies will be carried out in the defined fire blight environment of EPPO field trials. Cultivable bacteria will be monitored in parallel to the control treatments via MALDI-TOF MS ID. We will develop a suitable protocol for population monitoring and compare results with other PHYTFIRE partners. Population monitoring will allow for early detection of population shifts caused by fire blight treatments. It will contribute to enhanced fire blight prognosis and targeted application of antagonists.
