Purpose of research: Applied research

The aim of the project is the identification of fire blight resistant wild species of Malus as basic material for further breeding processes. The resistance of the Malus wild species M. ×robusta 5 was characterized by artificial shoot inoculation with different isolates of Erwinia amylovora. The deletion of the avrRPT2Ea effector gene of the isolate Ea1189 was able to break the resistance of M. ×robusta 5, whereas 4 wild species accessions (M. fusca, M. floribunda, M. ×robusta ‚Persicifolia' und M. baccata) remain resistant. Therefore, the 4 fire blight resistant Malus wild species will be artificially inoculated in the glasshouse with other effector mutants of E. amylovora to identify resistance donors for further breeding (1). In parallel, an effector mutant collection will be established. Additionally, resistant Malus wild species will be inoculated with the E. amylovora isolate Ea3049 which is also able to break the resistance of Mr5, to identify other resistance mechanisms. Furthermore, the migration of E. amylovora through grafted resistant wild species accessions into the rootstock shall be investigated. The selected wild species accessions with different resistance mechanisms will be identified as basic material for future apple resistance breeding against fire blight. The project supports the strategy of the ministry to combat fire blight without antibiotics and contributes to the National Action Plan on the Sustainable Use of Plant Protection Products to further reduce the risks of man, animals and the environment.
