Purpose of research: Applied research

The aim of the overall project is to predict the phenotypic response of barley genotypes to drought stress in different environments by integration of genetic information (QTLs) into physiological models of growth. Drought stress is one of the most important yield limiting factors all over the world as well as in numerous barley growing areas in Germany. The aim of the subproject 'Identification of genes and indicators for drought stress tolerance in barley (Hordeum vulgare)' is to identify QTL for drought stress tolerance via associations mapping. For this purpose, 192 genetically diverse spring barley genotypes are genotyped with a high number of SNP markers. In addition to this genome-wide approach, candidate genes for drought stress tolerance, which are known from literature, are included into the association studies. Furthermore, the extreme genotypes of the population are analyzed by Real-Time PCR in order to detect genotypic differences related to the expression of genes for drought stress tolerance. Phenotyping is done by the project partners with regard to the response to drought stress in the vegetative and the generative developmental stage. The phenotypic and genotypic data generated are the basis for the identification of QTL for drought stress tolerance in barley via association mapping. The QTL detected as well as the phenotypic and genotypic data will be used for the modeling. It is expected that this work will contribute to the understanding of the genetics of drought stress tolerance in barley. Based on these findings it should be possible to breed for drought stress tolerance and to model yield development under drought stress. As a preliminary work for this project, the spring barley genotypes were propagated in Groß Lüsewitz during the growing season 2010. Furthermore, the set of genotypes has been genotyped with 1536 Illumina markers, 1935 DArT markers and the 9kiSelect chip previously. These data are available for the current project. In the subproject 'Identification of genes and indicators for drought stress tolerance in barley (Hordeum vulgare)' 189 genetically diverse spring barley genotypes were genotyped by Genotyping-by-Sequencing (GBS) in collaboration with the IPK Gatersleben. For this purpose, a GBS-library was constructed and sequenced on an Illumina HiSeq machine (1 lane, 1 x 100 cycles) via Sequencing by Synthesis. The next step is the analysis of the sequence data (20 GB). It is expected that in this way a sufficient number of markers could be generated in order to perform a genome-wide association study.
