Purpose of research: Basic research

The work will be carried out by a bilateral project team comprising internationally-renowned academic and industry partners from Russia and Germany, thus ensuring the provision of high-quality data that will facilitate the translation of novel T. koksaghyz lines, extraction technologies and knowhow to the market in the form of high-quality rubber products. WP3 (JKI): Generation of TILLING and TALE nuclease populations of T. koksaghyz plants. We will use disarmed Agrobacterium tumefaciens strains for the transient expression of site directed nucleases (SDNs), presented as non-integrating T-DNA vectors, in T. koksaghyz leaf tissues. The explants will be regenerated under imidazolinone selection to identify specific point mutations introduced into the AHAS gene. Natural T. koksaghyz populations and those created by TILLING and using TALENs will be screened initially under greenhouse conditions for imidazolinone resistance. The most promising plants will be used to produce seeds for further screening under field conditions in parallel trials conducted in Germany and Russia. The performance and sustainability of imidazolinone-resistant plants will be monitored in parallel in Germany and Russia.
