Purpose of research: Applied research

Changes in the plant metabolom in consequence of heavy metal contamination in the soil should be evaluated for the model organisms oat (Avena sativa), beet (Brassica rapa L. var. rapa) and basil (Ocimum basilicum) for highly copper charged soil. Therefore, the main metabolic profile of fresh and dried/milled plant material should be analyzed by HPLC, GC and FT-IR and Raman-spectroscopy methods. Additionally, the fatty acid profile of plant cell membranes will be evaluated by extraction, saponification and subsequent GC-FID analysis and GC-library evaluation (Microbial Identification System-Software) to visualize changes in reaction to increased copper loadings in the soil. Thereby, if a standardization of this method according to DIN/ISO certification is possible, this method will serve as additional biological methodology for characterization of soil quality. Furthermore, both analytical approaches will be used on the one hand for estimation of vibrational spectroscopy as tool for analyzing the FA profile in plants and on the other hand for the evaluation of changes in the plant metabolic profile in general. The subsequent analysis of plant extracts for exact determination of changing metabolites by LC-MS will be aspired after establishment of the HPLC/LC-MA working group in spring/summer 2014.
