Purpose of research: Applied research

Due to a concentration range of a few femtogram, studies concerning dioxins and PCBs in food need high standards for the analysis. Commission Regulation (EU) No 252/2012 lays down methods of sampling and analysis for the official control of levels of dioxins, dioxin- like PCBs and non-dioxin-like PCBs in certain foodstuffs. However, detailed information on sample preparation, especially fat extraction and clean up are missed. Due to the absence of information the official control laboratories have developed their own methods for these steps. The source of dioxin- and PCB exposure of humans are mainly foods of animal origin. For specification of the content of dioxins and PCBs fat is almost exclusively used as reference (pg WHO-TEQ/g fat). There is a high diversity of fat components in food. Due to the chemical composition of the fat, polar and non-polar components can be differentiated. At the fat extraction, the polarity of the solvent as well as the extraction method can influence type and amount of extracted fat. That’s the reason, that the results of analyses for the same foods can vary depending on the used solvent and method. This allows different decisions about exceedances of maximum levels in food. In order to achieve comparable results at the EU level, the standardization and harmonization of fat extraction at the analysis of dioxins and PCBs is essential. Aims of this project: 1. Studies about the effect of solvent and extraction method on type and amount of extracted fat from the matrices meat, fish, egg and milk powder. 2. Analysis of the components of the extracted fat respectively for conclusions on the polarity of the ideal solvent for the matrices 3. Studies about the effect of solvents and extraction methods on the amount of extracted dioxins and PCBs in grown and spiked samples
