Purpose of research: Basic research

The objectives of this 28-day oral toxicity study in rats with 2-monochloro-1,2-propane (2-MCPD) diester and unbound 2-MCPD / 3-MCPD were i.) to examine liver, kidneys and testes by traditional histopathology and ii.) to investigate in parallel the usefulness of proteomics analysis. The dosing scheme was based on Schilter et al. (2011) for a 28-day oral test in rats. To induce detectable effects a dose of 10 mg/kg/day 2-MCPD and equimolar doses of 2-MCPD diester (= 53 mg/kg/day - high dose or 13.3 mg/kg/day - low dose) were administered. One day following the last treatment liver, kidneys and testes were prepared for histopathological examination and for proteomics analysis (the latter done by BfR). Results: Absolute and relative wet weights of kidneys and liver upon necropsy of rats were increased in the 2-MCPD and the 3-MCPD dose group (statistically not significant). Histopathological examination showed effects in testes only; some changes (statistically not significant) were considered to be most likely compound-related. These effects are promising regarding the outcome of the concurrently running proteomics analyses.
