Major objective of project B6 is to unravel determinants in maize metabolism for the compatibility with the biotrophic fungus Ustilago maydis and the hemibiotrophic fungus Colletotrichum graminicola. Our aim is to uncover the function of genes that are essentially participating in the redirection of host metabolism in response to infection by respective pathogens. In both pathosystems, we employ fungal mutants with reduced virulence provided by our partners A1, A3 (C. graminicola) and A2 (U. maydis) for the study of the host metabolome and transcriptome during the interactions to pinpoint metabolic routes in the host that are manipulated by the pathogens. In project A1, a complementary functional characterization of the C. graminicola mutant strains employed to study the host response in the project proposed here will be conducted. Using our transcriptome platform and metabolite analysis pipeline and employing data management and central analysis by project B7, we will study refined aspects of the C. graminicola transcriptome on planta (A1 and A3) and the transcriptome and metabolome in (a) leaves of transgenic barley plants deficient in RBOH (B9) and (b) roots of barley gibberellic acid (GA) signalling mutants (B2) during the interaction with the mutualistic fungal endophyte Piriformospora indica in projects led by the indicated partners.
