Determination of coumarin in different food samples (Project)

...From the European Food Safety Authority (EFSA) a Tolerable Daily Intake (TDI) of 0.1 mg/kg body weight was established. In Germany during the last years in food surveillance high concentrations of coumarin (up to 10g/kg) in cinnamon were measured. ...To enable also the analysis of samples with lower coumarin concentrations existing methods have to be optimised in regard to limit of detection. The developed method(s) should be validated by different validation concepts. With the developed method the concentrations of coumarin in different cinnamon containing food samples (e.g. tea, other beverages) and the influence of their preparation on the coumarin concentration will be determined. The concentrations of coumarin in other samples should be investigated. Since only some data on the bioavailability of coumarin from the different food materials are available in an animal experiment with a small number of animals the bioavailability of coumarin from coumarin containing products will be investigated....

Migration of nanosilver and nanoclay from food packaging materials into food (Project)

...For this purpose a masterbatch with LDPE (ExxonMobil™ LDPE LD 100) as polymer basis, which contains AgPURE™ nanosilver (rent a scientist GmbH, D99 under 20nm), was extruded to films with a thickness of 40 µm and a nanosilver content in the range of 500 ppm to ca. 11000 ppm (film 3 - 7). A film made of LDPE without nanosilver was produced as blank test material. ...The highest silver release was determined to be 6.8 ± 0.8 µg/dm2 with film 7 and 3% acetic acid and the lowest silver release was observed in the commercially available films in trace concentrations ranging from 3 ± 1 ng/dm2 to 33 ± 3 ng/dm2. ...For the test series the fillets were packed in nanosilver containing bags (blank and film 14) and for reference in sterile whirl-packs and stored at 4°C in refrigerator for 7 days. 10 colonies were randomly picked for bacterial diagnostics to check, whether the bacterial flora have changed in composition due to different storage conditions. ...

Molecular effects of flavonoid quercetin on human primary hepatocytes compared to those in mouse liver (Project)

...Due to its predicted anti-inflammatory, anti-oxidative and anti-carcinogenic properties food supplements with a daily intake up to 2 g were recommended. However, the effects of such a high dose on humans can not be assessed. An in vivo study in mice revealed that orally given quercetin (10 mg/kg body weight for 3 days) causes a drastic change in mRNA pattern in the liver of fed mice compared to control animals. 1790 genes were identified to be differentially regulated with a fold change of 2 and a q-value of 0.05. In this study human primary hepatocytes should be used to analyse the molecular effects of quercetin on human liver The liver hepatocytes should be exposed to 3 different human relevant doses of quercetin covering the normal intake up to the dose ingested by supplements. ...
