Purpose of research: Applied research

Generation and characterization of transgenic animals require great efforts. The population of those mutant lines is most often very small, yet their scientific benefit is enormous. Following generation and characterization, transgenic lines must be kept in stock, even if they are out of any experimental use. Otherwise they will be lost and can be recovered only hardly.Needs to save space (and money) for mouse housing, to rederivate infected mouse lines, and the increasing problems to import or export mice made a search for alternatives to standard breeding procedures mandatory: As an alternative to maintain (mutant) lines in stock and to protect them against an unexpected loss.The cryopreservation of early embryonic stages is a powerful tool to save and to handle the increasing number of transgenic mouse lines. Cryopreservation of transgenic mice is being done at the German Cancer Research Centre (DKFZ) in Heidelberg since ten years. However, the outcome of embryos varies from line to line up to the factor of 10 and becomes a limiting parameter of the whole procedure. Using the data of 95.000 cryopreserved embryos the reasons for these variances were elucidated and strategies to omit the resulting consequences were developed. In addition possibilities to raise the efficiency were investigated.
