Purpose of research: Experimental development
Serologically, influenza viruses are classified into 16 antigenic subtypes of the surface protein haemagglutinin (HA 1-16) and 9 antigenic neuraminidase subtypes (N1-N9). These can reassort freely in addition to the other six segments of the influenza virus and can thus generate new viruses. In poultry, the subtypes H5 and H7 have so far been able to mutate to highly pathogenic variants. Currently, a differentiation of the individual HA-specific antibodies is only possible by the haemagglutination inhibition test (HI test). The test requires the continuous supply of poultry erythrocytes and the corresponding specific antigens (H1-16) and antisera (anti-H1-16). The aim of the project is to generate monoclonal antibodies, which permit a serological classification into a subtype by means of a species-independent ELISA procedure. Monoclonal antibodies H1-16 and N1-9 can also be used for automated subtyping (e.g. wildlife monitoring) or for the development of a biochip.
