Purpose of research: Applied research
Tumor promotion by phenobarbital or phenobarbital-type agents such as PCB153 consists in a positive selection for hepatocytes with activating mutations in the Beta-catenin gene (ctnnb1). The underlying mechanism is unknown. Amongst others, Beta-catenin is a transcrip-tional regulator partly involved in the regulation of gene expression in liver. The analysis of the β-catenin-controlled proteome patterns in liver and its modification by phenobarbital may give insight into the mechanisms underlying selection by the tumor promoter. It is planned to use phenobarbital-treated and untreated knockout mice with liver-specific elimination of Beta-catenin and respective wildtype mice to identify genes regulated by the oncoprotein and the tumor promoter.
