Purpose of research: Applied research
Aim of the project was to establish a vector based method of stable transformation of mammalian cells with siRNAs. This includes the optimization of vector design (suitable regions of the target gene, design of the siRNA insert), the optimization of the transformation efficiency and the development of methods showing the successful knock down of the target mRNA. Using this method the function of human proteins of the Myc-Miz-1-TopBP1-network should be investigated as an alternative of the use of knock-out mice.
