Drought stress like other stress factors induces diverse changes in the metabolism of plants. Its impact depends on the time, duration and intensity of the stress phases. Among others, plants respond to drought stress with the accumulation of different substances in the leaves, which may be used as indicators for drought tolerance.
At the Institute of Abiotic Stress Tolerance the evaluation of such specific drought induced traits is performed under controlled conditions (rain-out-shelter, in vitro culture). The identification of relevant indicators is the prerequisite for association studies in a large range of potato genetic resources.
While segregation analysis, the most frequently applied method for the identification of molecular markers or genes, is based on segregating populations from crosses between two different genotypes, the method of association mapping permits the utilization of existing biodiversity (recent varieties, old varieties, landraces) for the search of markers or genes, without the necessity of generating new segregating populations.
The aim of this project is to establish basic methods and techniques which facilitate the development of molecular markers for the drought tolerance of potatoes by the use of association mapping with the aid of candidate genes from sequence databases.
