Purpose of research: Applied research

The purpose of the project is to screen for a possible teratogenic potential of certain flavour-ing substances (e.g. 2-methyl-2-propenoic acid, 2-methyl-2-butenoic acid and 2-methyl-2-pentenoic acid) and natural food ingredients structurally related to the known teratogen val-proic acid. In-vitro-methods such as cell culture, reporter gene assays and Western blot have been used. Inhibition of histone deacetylases as analysed by Western blot and F9 cell differentiation as investigated in a reporter gene assay were used as indicators for a terato-genic potential. It is planned to repeat single experiments and to test these substances in a third in vitro assay, the embryonic stem cell test. Results could become relevant for the risk assessment of flavourings by EFSA in the framework of the implementation of a positive list for flavourings in the EU.
