The aim of this project is to develop a pipeline for an efficient and transgene-free gene targeting in tomato plants, allowing the introduction of targeted mutations in the genome, the exchange of regulatory regions or the replacement of isogenes, for the improvement of plant traits, such as an increase of anthocyanin content in fruits. Currently, gene targeting is possible but occurs at low frequency and requires the use of selection markers, which is not desirable due to their transgenic character. We will combine innovations 1) in the marker-free detection of gene targeting event, 2) in increasing the frequency of gene targeting events, 3) in the improvement of plant regeneration methods and 4) for the establishment of regeneration protocols for tomato varieties used commercially. Although the focus of the project is on tomato, it is expected that the technologies developed during the project will be applicable to other crops.
