Purpose of research: Experimental development

The goal of the project is to develop and establish a new technique for the ribonucleoprotein (RNP) introduction and selection of the events to create non-GM abiotic stress like cold, drought, and salinity-tolerant crop plants within a short period of time. We want to develop a new method called the "cavitation bubble-induced shock wave-mediated RNP introduction" into plant cells that might cause less damage to the cells, and therefore result in high regeneration efficiency of the mutated plants. When using RNP, no marker genes are introduced into the genome and therefore screening for mutations is laborious and time consuming. In this project, we will attempt to solve this problem using a fluorescent dye attached to RNP, and then select cells that have fluorescent RNP using a micromanipulator. Further screen the mutated plants using high resolution Melting-Curve analysis for simple and fast detection of the mutation, and confirmed by Sanger sequencing. The selected plants will be grown in the greenhouse for the development of T2 homozygous mutants. The developed T2 homozygous plants will be further subjected to abiotic stress tolerance analysis to identify the best genotypes.
