Effects of the endogenous pectate lyase (PL) 1 on the induction of plant defence mechanisms in transgenic potatoes (Solanum tuberosum L.)

Purpose of research: Basic research
Transgenic potato lines of cv. Dèsirèe expressing the isoenzyme PL1 of Erwinia carotovora will be generated. The PL1 transgenic plant lines will be grown in the greenhouse and the tubers will be examined with respect to properties of relevance for host resistance against Erwinia soft rot. It is assumed, that the isoenzyme PL1 has an enhanced capacity to induce plant defence mechanisms due to its ability to release longer chained, i.e. hepta- and pentamer unsaturated oligoglacturonates (OG) from plant cell wall pectin. For this reason the effect of PL1 expression on tissue cell walls, polyphenol oxidase (PPO), phenylalanine ammonia-lyase (PAL) and peroxidase (POD) as well as lignin and suberin, marker for an induction of plant defence mechanisms, in transgenic potatoes will be examined.
The project is useful to draw up parameter for the evaluation of plant defence mechanisms. It is assigned to safety research.
