Loose smut is one of the most important seed born diseases in barley. There are strict limitations in the percentage of infected plants in a seed multiplication field. In spite of increasing production of beverages made from barley or malt and in particular a still growing market for organic beer is still growing, it is of elementary interest to provide varieties wich are well adapted to the conditions of organic farming. This means that the varites should not only show good agronomic performance under low input conditions but also provide tolerance against seed born diseases as well as good malting quality. A number of diffenet sources for resistance against loose smut is known but it is not yet clear if they are different alleles of one single locus or independent genes. To obtain more information abot the characteristic od these resistances istis necessary to analyse the genetic segregation of the tolerance in different genetic backgrounds. Therefore a number of different crosses has to be analysed for the presence of resistance. Two generations are required to identify resistant plants in the reproducible greenhouse test established at the LfL. Therefore it would be very beneficial to develop molecular markers for the selection of tolerant breeding lines early during the breeding process. In addition a reliable method for the quantification of the fungal infection will be a tool to identify infected plants. The proposed project has the intention to identify different resistances against loose smut and to develop molecular markers for selection and a qPCR assay to quantify the infection with this dangerous fungus.
