The aim of this study is the development of novel and innovative analytical approaches for the mass spectrometry-based detection of allergens in food. Food allergens are one of the major food safety concerns in industrialized countries and specific allergen detection methods are key to efficient allergen management. Currently, specific and sensitive methods for the reliable quantitation of allergens are not available in routine laboratories.We therefore aim to develop an automated multi-allergen method for the detection and quantitation of relevant allergens using peptide biomarkers as analytes (targeted proteomics). As necessary limits of detection are extremely low for some allergens, we in addition aim to provide affinity-based enrichment strategies to enhance sensitivity and specificity. The mass spectrometry-based detection of food allergens is a very promising approach. However, for the application in routine laboratories, further research and development is necessary. The current technological gaps will be subject of this project to allow the i) systematic identification and optimization of critical parameters of sample preparation ii) automization of sample praparation to enable routine laboratories without prior knowledge in proteomics the applicatiopn of MS-based allergen detection iii) Development of affinity media to allow the use of current MS infrastructure in routine laboratories for allergen detection (enhancement of sensitivity) iv) Development of a multi-allergen detection method that covers all relevant food allergens v) Development of a fast screening method and a method for reliable quantitation.
