Food and consumer protection

Project code: MRI-MBT-08-1040 Bakterien Nigeria-Deutschland
Contract period: 01.03.2017 - 28.02.2018
Purpose of research: Applied research

The studies aim to determine and compare the sources and incidences of antibiotic-resistant bacteria and distribution of resistance genes in Staphylococcus (methicillin-resistant Staphylococcus aureus-MRSA), Escherichia coli (ESBL-Escherichia coli, E. coil O157:H7 and other pathotypes and Salmonella species such as Salmonella Enteritidis and Salmonella Typhimurium), as well as their specific lineages in plant and meat products from food chains in Germany and Nigeria. The study furthermore aims to genotypically identify antibiotic resistant bacteria and where possible to characterize the resistance genes by whole genome sequencing.

To date, approx.. 400 bacteria were isolated from fresh produce and fresh meat products and animals and these were phenotypically characterized in order to presumptively identify the bacteria. The antibiotic resistances of the 400 strains were determined by disc diffucion assays. Of the 400 strains, about 100 were investigated in more detail and these were identified using genetic characterization techniques including 16 S rRNA gene sequencing and PCR typing. The genomes of 25 of these strains from plant foods were completely sequenced. The first results have recently been published: Igbinosa, E., Rathje, J., Habermann, D., Brinks,, E., Cho, G.-S., Franz, C.M.A.P. 2018. Draft genome sequence of multidrug-resistant strain Citrobacter portucalensis MBTC-1222, isolated from Uziza (Piper guineense) leaves in Nigeria. Genome Announcements 6(9), e00123-18.
