Within the project PhytoSica it is intended to develop a rapid detection method based on immunological and molecular procedures for three important European fruit phytoplasmas, namely for the quarantine pathogen Candidatus Phytoplasma vitis and for the regulated non-quarantine pathogens Candidatus Phytoplasma mali and Candidatus Phytoplasma pyri. For each pathogen a specific monoclonal antibody will be developed which is directed against the frequently occurring membrane protein IMP, or VMP for Ca. P. vitis. The monoclonal antibodies are linked to reaction vial surfaces and will bind phytoplasma cells from plant extracts. Subsequently, the same antibody, now conjugated with DNA-fragments, is added and will bind to free epitopes. In the following step the conjugated DNA-fragment will serve as the target for the isothermal DNA-amplification by a recombinase. The enzyme will incorporate modified primers which can be visualized colorimetrically on a dipstick after the reaction. The whole procedure is performed with simple devices and suited for a point-of-care application.
