Purpose of research: Experimental development

A DH population derived from a cross between the winter barley cultivars Igri and Franka was used for genetic mapping of QTLs associated with resistance to the root lesion nematodes caused by Pratylenchus neglectus and Pratylenchus penetrans. Artificial inoculation with P. neglectus and P. penetrans was carried out in greenhouse and climate chamber experiments. Continuous phenotypic variations in infection rate were observed for both nematode species indicating a quantitative inheritance of resistance. An existing genetic map was enriched by 527 DArT markers. The new genetic linkage map comprised 857 molecular markers that cover 1,157 cM on seven linkage groups. Using phenotypic data, five QTLs associated with resistance to P. neglectus were mapped by composite interval mapping on four (3H, 5H, 6H and 7H) linkage groups. Four QTLs associated with resistance to P. penetrans designated as Ppe2H, Ppe5H-1, Ppe6H-1 and Ppe7H were mapped on linkage groups 2H, 5H, 6H and 7H, respectively. Comparative QTL analysis revealed a co-localization of resistance-associated QTLs of P. neglectus (chromosomes 5H Pne5H and 6H Pne6H) and P. penetrans Ppe5H-1 and Ppe6H-1.
