Purpose of research: Basic research

Nitrogen retranslocation from senescing leaves to the grains is one of the limiting factors for an efficient recovery of fertilizer nitrogen supplied to crop plants. Although molecular and physiological approaches have described enzymes, transporters and nitrogenous compounds available for nitrogen retranslocation, experimental evidence on the significance and efficiency of different nitrogen forms for retranslocation processes is poor. Based on our previous studies which have demonstrated the contribution of ammonium and urea transporters for nitrogen uptake in roots, the present project aims at elucidating their role in nitrogen retranslocation. First, the cell type-specific expression of the ammonium and urea transporters will be investigated in senescing leaves. Transgenic Arabidopsis lines with de-regulated expression of leaf-expressed transporters for ammonium, urea and amino acids will then be employed for short- and long-term translocation studies of 15N-labelled nitrogenous substrates. Phloem loading and retranslocation rates will be compared between leaves that are senescent due to nitrogen deficiency or due to plant age. Finally, senescence-dependent translocation processes of selected nitrogenous substrates will be verified in wildtype and transgenic barley lines.
