Purpose of research: Basic research
High yield and grain quality can only be achieved at optimum supply of the growing seed with carbon and nitrogen assimilates. In barley the ear-near photosynthesising organs, flag leaf, glumes and awn, are most important for supplying the growing grain. During plant senescence C and N compounds are mobilised from these organs and transported to the filling grain. Therefore, understanding the mechanisms of sink-induced senescence and nutrient remobilisation can potentially contribute to the generation of improved crops with higher N-use efficiency.
This project addresses interactions between senescence-induced N remobilisation and seed filling in barley. It is planned to study the relationship between senescence and seed filling using profiling of gene expression, enzyme activity and metabolites.
Analysis will be done in parallel, in the vegetative ear-near organs, flag leaf, glumes-fraction and in the developing grain. Objectives are:
i) to understand the influence of seed filling on senescence and C/N remobilisation in flag leaves and the glumes fraction (awn, glume, lemma, palea) and
ii) to understand the effect of increased seed sink strength, achieved by transgenesis, on senescence development and N remobilisation.
In a second step N-transporters involved in senescence induced N-remobilisation will be identified and characterised. A barley transformation platform will be established to validate putative candidate transporters and as service for the research group.
