Purpose of research: Experimental development

A reliable and sensitive procedure was established to produce double-stranded DNA from genomic RNA of avian influenza viruses. The double-stranded DNA is required to conduct sequencing processes with the Genome Sequencer FLX. To handle the accumulated and associated data from sequencing, a database was created in the program R, which enables controlled and effective data manipulation and evaluation. By implementing and considering relevant crucial operations and variables in the database, reliable results are highly expectable. To comply with the requirements for a selective analysis of avian influenza virus-sequences, specifically adapted processes were programmed for the relevant genome. These processes enable an optimized assembly, a valuation of variability of virus populations, a determination of open reading frames and encoded proteins and a search for closely related viruses. Furthermore, in concordance with the established procedure a process was prepared to analyze changes of viral quasi-species, which potentially occur from interspecies transmission processes. In conclusion, an applicable method was generated, which allows a rapid and detailed analysis of entire genomes.
