Purpose of research: Basic research
It is hypothesized that MAP isolates with different genotype have also different phenotypic properties which contribute to the variability in the clinical outcome of MAP infection. The present project will contribute to the clarification of this issue. The objectives of the project are: 1) Detailed characterization of representative MAP isolates, which have already been genotyped in a previous study, by detection of Large Sequence Polymorphisms (LSP´s), Multi Locus Sequence-Typing (MLST) and using a pan-genomic micro-array in order to clarify the phylogenetic development of the isolates. 2) Phenotypic characterization of genotypic different MAP strains in vitro by studying their interaction with THP1 macrophages and human and bovine intestinal epithelial cells. The influence of milk as a hyperosmolar medium on invasion of the strains will also be studied.
