Purpose of research: Basic research

There is a wide spectrum of obesity-associated pathologies, including hypertension, dyslipidemia, glucose intolerance and systemic low-grade inflammation. Results of our own studies clearly show that distinct foods (e.g. apple) contain ingredients which could cure or delay the progression of obesity related complications such as the systemic and hepatic dysregulation of Lipid homeostasis accompanied by chronic inflammation. The aim of our work is to identify the bioactive components in food and further characterize hepatic and adipose tissue pathways which are responsible for observed anti-steatotic and antiinflammatory actions. For aiming this, combined in vitro and in vivo strategies help identifying bioactive food ingredient(s) with focus on mechanistical evaluation of relevant (lipogenic, lipolytic, (anti-)inflammatory) gene expression profiles and pathways also in context with ´activity guided fractionation´ strategies and application of monosubstances. By further using established animal models for obesity mimicking hepatic and systemic pathologies of human metabolic syndrome, the in vivo bioactivity of identified food ingredient(s) could be confirmed and further approved in frame of human intervention trials. As this concept includes cellular and in vivo models based on different species the expected results will identify novel candidates with relevance for preventive dietary strategies in human nutrition and possibly novel perceptions on the underlying mechanisms of their action.
