Purpose of research: Applied research
Eyespot on wheat, caused by Pseudocercosporella herpotrichoides (Fron) Deighton, is the most important stembase disease of wheat in humid and cold regions. The frequency of its occurrence and the diseae severity is closely related to the cropping system. Because the resistance against P. herpotrichoides has a small to medium heritability, only, the selection of resistant genotypes is very difficult and the availability of molecular markers will be advantageous. A precondition for the selection of resistant genotypes as well as for phenotyping of doubled haploid lines (DH-Lines) for the development of molecular markers is the establishment of an efficient test for resistance. The accuracy of the resistance evaluation is essential for the development of molecular markers and finally their application in practical breeding. Based on the development of an effective test system closely linked markers for the resistance genes Pch1, Pch2 and the Ae. kotschyi resistance using DH-lines and the ‘Bulked Segregant analysis’ (BSA) will be developed. Using these markers a combination of all resistance genes can be achieved leading to a considerable improvement of the level of resistance against P. herpotrichoides.
