"Livestock harbour a wide variety of DNA viruses. This project aims at comprehensively elucidating the incidence of viral DNA especially in cattle herds and food of animal origin. With respect to preventive consumer protection and animal welfare, the project will 1. Determine the incidence of viral DNA in cattle and food made from cattle, 2. Eventually identify new DNA viruses in cattle herds, 3. Collect evidence of the potential pathogenicity of new or less characterized viruses. In this effort, three partners cooperatively address the different questions in 9 work packages. The partners are LMU München (Lehrstuhl für Virologie; Lehrstuhl für Hygiene und Technologie der Milch), die CAU Kiel (Institut für Tierzucht und Tierhaltung) und das Friedrich-Loeffler-Institut (Institut für Virusdiagnostik). The tasks of the work packages are AP1 Rolling circle amplification of circular DNA extracted from cattle samples (CAU, LMU-V), AP2 Metagenomic sequencing of cattle samples (German Simmental, Holstein-Frisian) both from DNA and RNA (FLI), AP3 Bioinformatic analyses of datasets generated in AP2 and comparison with data available from analyses previously conducted at FLI (FLI), AP4 Identification of potential target sequences and viruses in the results gained from previous work packages (CAU, FLI, LMU-V), AP5 Development of tools for the characterization of viral proteins with monoclonal antibodies generated using recombinantly expressed proteins (LMU-M), AP6 Targeted detection of target-sequences in samples from different species (LMU-V) and different foods of animal origin (CAU), respectively, aiming at the prevalence comparison, AP7 Antibody-based detection of target antigens (LMU-M), AP8 Functional analyses of relevant new or yet uncharacterized viruses in vitro or, if appropriate, in vivo (LMU-V), AP9 Compilation of relevant knowledge for risk assessment and definition of consequences (CAU, FLI, LMU-M, LMU-V)".
