Occurence of Pasteuria penetrans in field populations of Heterodera schachtii in soils with intensive rotation systems of oilseed rape and sugar beets and investigation on pathogenicity of different isolates

The beet cyst nematode Heterodera schachtii belongs to the main pests of sugar beets and is largely established in growing regions in Germany. Numerous natural antagonists occur in association with nematodes, which could successfully been applied as biological control agents. Biological control methods in soil that make use of micro-organisms show a reasonable efficiency at higher soil temperatures, because organisms previously were isolated an selected in warmer growing regions. The proposed project aims at the nationwide survey of the natural occurring obligate endoparasite Pasteuria penetrans in field-populations of the beet cyst nematode. Due to the establishment of efficient multiplication techniques of certain life stages of the endoparasite it is possible to excess infection material for controlled experiments in short term. Using this material, pathogenicity of different P. penetrans isolates against H. schachtii should be studied. Another objective of the project is to find isolates which show a high potential for the development of biological control method against beet cyst nematodes for central European beet growing regions.
Pathogenicity of a Pasteuria nishizhawae isolate towards H. schachtii in susceptible and tolerant sugar beet genotypes were testes in greenhouse experiments. The isolate was provided by Syngenta Crop Protection. Additional test aimed at the successive effects of repetitive inoculation of Pasteuria spores on Beet Cyst Nematodes (BCN) population dynamics in sugar beet pathosystem. Microplots were installed in 2014 and replanted with spore treated sugar beets in 2015 for a second rotation. In another experiment BCN population dynamics and sugar beet performance was observed after repetitive Pasteuria inoculation for two BCN generations under outdoor and two generation under greenhouse conditions. Effects of Pasteuria on sugar beets and BCN population dynamics were observed but could not be reproduced. Further investigations are needed to identify supportive and inhibiting factors in the Pasteuria-BCN-Sugar beet-System.
