Purpose of research: Applied research

The risk potential of inhalable particles, which are generated either unintentionally by combustion processes or in the production or application of industrially manufactured nano-particles, is determined almost exclusively in animal tests by inhalation or intratracheal instillation. To examine the biological effects of inhalable particles by in vitro test methods either collected particles are suspended in culture medium and brought into contact with suitable cell lines in submersed culture or the cells are exposed with aerosol at the air-liquid layer. The first method is suitable for a screening, however, it hardly corresponds to the physiological conditions during inhalation. The second method is experimentally more complex and is so far only little used. Therefore, a standardized in vitro test procedure with a lung-specific bioassay is developed for the toxicological evaluation of aerosols.
